NSC from mouse brain can be differentiated into various neural and glial cell types, potentially in the future this may contribute to the development of therapies for CNS disorders via replacement of damaged neural tissue. The Nucleofector technology presents a powerful tool to gain further insights into how these cells function.

Nucleofection of mouse NSCs
Mouse neural stem cells were isolated from spinal cords of mouse embryos (E14-15) and cultured for 3 passages in medium containing EGF and FGF. Cells were transfected by nucleofection using program A33 and 3 µg of a plasmid encoding the enhanced green fluorescent protein eGFP. After nucleofection the cells were cultured in medium containing 1% FCS to initiate differentiation into neurons and astrocytes. 72 hours post nucleofection cells were analyzed by fluorescence microscopy.
