Macrophages are frequently used in studies on basic immunology and cancer research. With the Mouse Macrophage Nucleofector Kit you can now address areas such as gene regulation, signaling pathways or identification of genes responsible for differentiation in the murine in vitro model system.

Example for the nucleofection of mouse macrophages with pmaxGFP.
Primary mouse macrophages (isolated from C57BL/6 mice) were transfected by nucleofection with 2 µg pmaxGFP™ and analyzed by light (top) and fluorescence microscopy (bottom) at 40x magnification. Transfected macrophages reveal cytoplasmic extrusions important for phagocytic function of macrophages.
Mouse macrophages can be stimulated after nucleofection
Primary mouse macrophages (C57BL/6) were transfected with 2 µg pmaxGFP™. 6 hours post nucleofection the culture medium was changed and cells were stimulated with LPS. 24 hours post nucleofection TNFα production was determined by ELISA. Values were normalized to number of living cells after transfection and are expressed as pg/ml TNFα per 1000 cells. In DNA transfected samples a similar but slighlty reduced TNFα production can be detected. In non-stimulated samples no TNFα was detected.
