The capability of bone marrow derived MSC to differentiate into various cell types such as osteoblasts, adipocytes, chondrocytes, myocytes or tenocytes has led to great interest from many different research fields.
Mesenchymal stem cells have the potential to differentiate into cardiomyocytes, thus turning them into an interesting cell type for cardiovascular research.
Investigations of human MSC have so far been hampered by the fact that these cells could only be genetically modified using retroviral vector sytems. The Human MSC Nucleofector Kit now permits for the first time a non-viral and efficient transfection of these cells. With the Nucleofector technology, transient gene transfer into human mesenchymal stem cells can be achieved with up to 85% efficiency.
Nucleofection of human mesenchymal stem cells.
Human MSC (Human Mesenchymal Stem Cells) were transfected using the Human MSC Nucleofector Kit, program U-23 and a plasmid encoding the fluoresent protein eGFP. 21 hours post nucleofection the cells were analyzed by flow cytometry (FACS). eGFP expression of Human MSC is shown after nucleofection without (A) and with plasmid DNA (B).

Cells were transfected by nucleofection using program U-23 and 5 µg of a plasmid encoding the mouse MHC class I heavy chain molecule H-2Kk.
