Efficient transfection of human B cells is a pre-requisite to enable research in fields such as chemokine/antigen response or B cell activation. The standard Human B Cell Nucleofector Kit for 100 µl cuvettes has been successfully used for both DNA and RNA transfection and its applicability in many research areas has been demonstrated in various publications. With the new Human B Cell 96-well Nucleofector Kit it is now possible to perform nucleofection in 96-well format.

Transfection efficiency of freshly isolated human B cells 24 hours post nucleofection.
Freshly isolated human B cells were nucleofected with program 96-EO-117 and 0.4 µg of pmaxGFP. 24 h post nucleofection cells were analyzed by FACS. Cell viability is usually around 70% (%PI negative B cells) after 24h.
Uniform reporter gene expression of 96-well nucleofected human B cells
Freshly isolated human B cells were nucleofected with 0.4 µg pmaxGFP using the Human B Cell 96-well Nucleofector Kit. 24 hours post nucleofection cells were analyzed by flow cytometry. Wells 1 and 96 are negative controls of cells in 96-well Nucleofector Solution and plasmid DNA, but without nucleofection.
