Transfection of primary mouse hepatocytes represents a major breakthrough in gastroenterology research. Efficiencies of up to 60% can be achieved using nucleofection. The Nucleofector Kit for freshly isolated mouse hepatocytes is suitable for both DNA and siRNA transfer, while functionality of cells is not affected.

Nucleofection of primary mouse hepatocytes
Mouse hepatocytes were transfected with pmaxGFP. 24 hours post nucleofection cells were analyzed by light (A) and fluorescence microscopy (B).



Efficient gene silencing in mouse hepatocytes.
Hepatocytes were transfected by nucleofection with 2 μg of pmaxGFP™ (A,D), cotransfected with 2 μg pmaxGFP and 1.5 μg unspecific siRNA (B,E) or with 2 μg pmaxGFP and 1.5 μg siRNA directed against maxGFP™ (C,F). 24 hours post nucleofection cells were analyzed by light and fluorescence microscopy for maxGFP expression.


Nucleofected hepatocytes maintain functionality.
24 hours post plating nucleofected primary mouse hepatocytes and non-nucleofected control cultures were analyzed for albumin secretion (normalized to cell number) by ELISA. Transfected hepatocytes show albumin secretion rates comparable to those of untransfected control hepatocytes.
