Aortic smooth muscle cells are a well established cell system that enables studies on human vascular disorders, such as atherosclerosis and stroke. As a result of the slow proliferation rate of aortic smooth muscle cells the expression of a transiently transfected gene can be monitored for up to three weeks (see below).

Time course of transient expression of transfected human AoSMC.
Human AoSMC were transfected using the Human AoSMC Nucleofector Kit and a plasmid encoding the mouse MHC class I heavy chain molecule H-2Kk. 1, 2, 7, 14, 21 and 28 days post nucleofection the cells were analyzed for their H-2Kk expression by flow cytometry. Dead cells were excluded from the analysis by propidium iodide staining and gating.
