MEF display significant genotypic and phenotypic variations depending on the strain and the genetic background of the mice they were isolated from as well as the immortalization strategy used. Thus, it was impossible to transfect some MEF cell types as each could behave differently with respect to the optimal conditions for efficient transfection. amaxa has resolved this drawback by developing two MEF Nucleofector Kits (1 and 2) and an easy-to-use MEF Starter Nucleofector Kit.

The MEF Starter Nucleofector Kit is designed to determine of the ideal nucleofection conditions for each MEF cell type. This is done in a total of just 4 reactions using MEF 1 and MEF 2 Nucleofector Solution with two different programs. In the next step the appropriate MEF Nucleofector Kit (1 or 2) can be used to perform the experiments.

Nucleofection of primary MEF

Spontaneously immortalized mouse embryonic fibroblasts (strain:C57B6Jx129SV) were transfected using the MEF Nucleofector Kit 1 and a plasmid encoding the enhanced green fluorescent protein eGFP. 24 hours post nucleofection, the cells were analyzed by fluorescence microscopy.
