The Cell Line Optimization 96-well Nucleofector Kit is the ideal tool to conveniently and rapidly determine the optimal 96-well nucleofection condition of virtually any difficult-to-transfect cell line within one experiment. This kit is suitable both for optimizing nucleofection of plasmid DNA, as well as, siRNA oligonucleotides.

Optimization Strategy
Nucleofection conditions optimized with our standard protocol using pmaxGFP are equally well suited for other nucleic acid substrates. Thus, siRNA oligonucleotides can be transfected using the same conditions as with plasmid DNA. This has been confirmed by comparing conditions for plasmid DNA and labelled siRNA.
Nevertheless, if you are interested in siRNA only (and have an established read-out assay for measuring the knockdown effect) you can test a reduced set of programs as recommended in our "siRNA only" protocol. Such program reduction is possible because siRNA is transfected much easier than DNA. For example, from the original set of 31 programs tested in the standard protocol "for plasmid DNA and siRNA" you may find three programs working comparably well for siRNA but only one of these three may work with DNA (especially if viability is an issue with DNA). Hence, the program you select for siRNA following the reduced strategy might not be suitable for DNA.
Optimization for Plasmid DNA and siRNA
31 different programs plus one control are tested with the Cell Line 96-well Nucleofector Solutions SE, SF and SG in one 96-well Nucleocuvette plate. Once the best Cell Line Nucleofector Solution and program have been determined amaxa's Scientific Support Team would be pleased to assist improving the results even further.
Optimization for siRNA Only
Using a control siRNA and a targeting siRNA, 15 different programs (plus one no program control) are tested in combination with the Cell Line 96-well Nucleofector Solutions SE, SF and SG in one 96-well Nucleocuvette plate. Once the best Cell Line Nucleofector Solution and program have been determined amaxa's Scientific Support Team would be pleased to assist improving the results even further.
