Example of nucleofection for 293 (ATCC)
293 (ATCC) were nucleofected with a plasmid encoding the fluorescent protein eGFP. 24 hours post nucleofection, cells were analyzed by light (top) and fluorescence microscopy (bottom).
Example of nucleofection for 293 (DSMZ) cells with H-2Kk cDNA.
293 (DSMZ) cells were transfected using the Nucleofector Kit V and a plasmid encoding the mouse MHC class I heavy chain molecule H-2Kk. 6 hours post-nucleofection, the cells were stained with a PE-coupled antibody directed against H-2Kk and were analyzed by flow cytometry. 293 cells were gated according to forward/side scatter (A). Dead cells were excluded by staining with propidium iodide and gating (B). H-2Kk expression in 293 is shown post-nucleofection without (C) and with plasmid DNA (D).
