We only recommend the depletion method. For Example, the RosetteSep® Isolation Kit for Human Monocytes (StemCell Technologies, Cat. No. 15628/15668). The advantage of depletion is that the monocytes are left untouched by antibodies during the process. It is also very important that the supplied recovery media has been supplemented with Glutamine and FCS as stated in the optimized protocol.
