«I routinely culture neural stem cells as neurospheres. Can these be transfected by nucleofection®?»

Yes. Sacrifice one dish of cells to determine the cell number. Use that determination to determine the cell number for the neurospheres that need to be transfected. Then incubate the neurospheres in Nucleofector® solution with DNA for 5 minutes. This helps promote the diffusion of the DNA into the interior of the sphere. Then, proceed with nucleofection® as usual. However, be careful to not incubate the cells in the Nucleofector solution longer than the recommended 15-20 minutes.
