Yes. For the best viability of HUVECs after nucleofection we recommend using only fresh HUVEC medium e.g. EGM-2 Bulletkit (Clonetics). At amaxa, we re-freeze our media in 40mL aliquots and store it at -80C°. We recommend freezing the media in aliquots because repeated freeze/thaw cycles, even if only a few times, may damage the growth factors (hVEGF, hFGF, hIGF) in the media. It also makes the media last longer.We have not seen any problems with re-freezing the media once.
