Mouse dendritic cells (DC) play a central role in the mammalian immune system, thus understanding their functionality for example by loss and gain of function studies is of vital interest for research groups and industry. The two Mouse Dendritic Cell 96-well Nucleofector Kits for immature and mature DC´s allow high throughput transfection with a wide variety of substrates enabling scientists to perform these studies with much less time and labor input compared to before.

Transfection efficiency of immature mouse dendritic cells 24 hours post nucleofection
5 x 10⁴ cells were nucleofected with program 96-DK-100 (high efficiency) or 96-DJ-100 (high viability) using 0.4 µg pmaxGFP. Cells were analyzed 24 hours post nucleofection using a Becton Dickinson FACSCalibur with HTS option. Cell viability was approximately 74% for 96-DK-100 and 98% for 96-DJ-100 after 24 hours (compared to PI negative cells of the untreated control).

Transfection efficiency of mature mouse dendritic cells 24 hours post nucleofection
2.5 x 10⁴ cells were nucleofected with program 96-DN-107 using 0.4 µg pmaxGFP. Cells were analyzed 24 hours post nucleofection using a Becton Dickinson FACSCalibur with HTS option. Cell viability was approximately 85% after 24 hours (compared to PI negative cells of untreated control).

5 x 10⁴ cells were nucleofected with 0.4 µg of pmaxGFP using the mouse dendritic cell (immature) 96-well Nucleofector Kit. 24 hours post nucleofection cells were analyzed on a FACSCalibur with HTS option (n=72, SD = ± 11.2% from mean). Wells without GFP expression are negative controls (cells in the 96-well Nucleofector Solution and plasmid DNA but without nucleofection).

Wells without GFP expression are negative controls (cells in the 96-well Nucleofector Solution and plasmid DNA but without nucleofection).
