Efficient transfection of human B cells is a pre-requisite to enable research in fields such as chemokine/antigen response or B cell activation. Now cited in various publications, the Human B Cell Nucleofector Kit has been used for both DNA and RNA transfection and has proven its applicability in many research areas.
- For unstimulated and stimulated B cells
- Up to 40% transfection efficiency
- One protocol for DNA, siRNA, RNA
Example for nucleofection of CD19+ B cells with H-2Kk cDNA.
Fresh, unstimulated PBMC were transfected using Human B Cell Nucleofector Kit and a plasmid encoding the mouse MHC class I heavy chain molecule H-2Kk. 16 hours post nucleofection, the cells were stained for CD19 and H-2Kk. Lymphocytes were gated according to forward/side scatter (A). Dead cells and CD19- cells were excluded by staining with propidium iodide and gating (B, C). H-2Kk expression is shown post nucleofection without (D) and with plasmid DNA (E).
