Establish your siRNA experiments faster than ever before! The siRNA Test Kit is based on co-transfection of pmaxGFP and a siRNA duplex directed against maxGFP. Knock-down of maxGFP protein expression can be easily detected by fluorescence microscopy. The kit has already been evaluated in multiple cell types such as Jurkat, THP-1, U-937, NIH/3T3 or primary NHDF cells. It is not suitable for use in primary human blood cells. For these cells an > Optimized Protocol for down-regulation of endogenous genes such as CD2, CD4 and vimentin is available.

Example for nucleofection of NIH/3T3 cells with pmaxGFP and siRNA.
NIH/3T3 cells (ATTC) were transfected with 0.5, 1 or 2 µg of pmaxGFP only (top row) or 1.5 µg siRNA directed against max GFP, in additon (bottom row). Gene silencing of maxGFP expression was monitored by fluorescence microscopy, 24 hours post-nucleofection.

Example for nucleofection of NHDF-Neo with pmaxGFP and siRNA.
Neonatal human dermal fibroblasts (NHDF-Neo) were transfected either with 1 or 3 µg pmaxGFP alone (upper) or co-transfected with 1 or 3 µg pmaxGFP and 1.5 µg siRNA against maxGFP. 24 hours post nucleofection gene silencing was visualized by fluorescence microscopy.
