Hot Topic: Strategies for Reducing Off-target Effects

Off-target effects can distort silencing results with false positive phenotypes or complicate interpretation due to inconsistent results. Such effects can be induced by sequence-dependent silencing of unintended targets or by unspecific effects of dsRNA. Our collaboration partner Dharmacon, Inc. has shown that the use of high quality siRNA reagents can help reducing these effects.

       Product Information

Nucleofector 96-well Shuttle
Whether you are transfecting siRNA oligonucleotides, shRNA vectors or miRNA substrates into hard-to-transfect cell lines or primary cells:

Nucleofector Technology offers you highly efficient non-viral delivery for any RNAi substrate combined with high flexibility – from single samples to library screening.

       Application Note

Knockdown of maxGFP in Jurkat cells
Delivering siRNA reagents or libraries into cells that have been considered intractable to lipid reagent-based transfection methods:

A guideline for establishing siRNA experiments by combining Dharmacon's highly functional SMARTpool siRNA reagents with the amaxa Nucleofector 96-well Shuttle System - a unique and powerful method.
